copyright infringement

An image of my artwork was published and copies offered for sale without my permission.  I never officially copyrighted my painting and wonder what steps I need to take to protect my work.  If I don't officially defend my copyright, does this mean that I will lose it?

no, you dont lose your copyright if you dont defend it.

You need to first register your work if you plan on suing anyone, and since the infringement occurred before your registration, what you can ask for might be severely limited. You'll want to contact a lawyer for details if you are seriously thinking of a lawsuit before you do anything. If however you just want them to stop profiting from your work and it is an online organization, a DMCA takedown notice might suffice.
